Officials say the USS John S. McCain collided with the oil and chemical tanker Alnic MC on Monday morning.
The US Navy is searching for the missing sailors along with Singaporean authorities.
The officials say the John S. McCain sustained damage to its port side. They say the ship is sailing under its own power and is heading to a port.
The John S. McCain is part of the US 7th fleet based in Yokosuka, near Tokyo.
Earlier this month, it took part in a US “freedom of navigation” operation near a Chinese-built island in the disputed Spratly Islands in the South China Sea. It was heading to Singapore when the collision occurred.
In June, another Aegis destroyer in the 7th fleet, the USS Fitzgerald, collided with a container ship off Japan’s Pacific coast. Seven sailors died in the accident.
